Abstract: Objective: To discuss the medication rules and mechanism of Chinese medicinals for tubal infertility (TI) based on data mining and network pharmacology. Methods: The relevant literature on TI treated by Chinese medicinals were included by searching the China National Knowledge Infrastructure (CNKI),China Science Periodical Database( WF),China Science and Technology Journal Database( VIP),PubMed and other databases from January 1st,2015 to September 1st,2023. The nature,flavors and meridian entries were counted by Excel software; the use frequency of Chinese medicinals was counted by R language and the top 20 Chinese medicinals were given association rules analysis and cluster analysis. The core prescriptions based on cluster analysis were obtained and the effective ingredients and targets were worked out by using the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP). The targets were corrected by UniPort database, the TI-related disease genes were researched on GeneCards database and Online Mendelian Inheritance in Man (OMIM) database, and the common genes were figured out by intersection analysis. Using Cytoscape 3.10.0,the protein-protein interaction (PPI) networks of core targets were obtained. The medicine-disease intersection targets were imported into Metascape for enrichment analysis of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). The Chinese medicinals-component-target-signaling pathway networks were established by Cytoscape 3.10.0 software. Results:A total of 120 prescriptions were included, involving 262 Chinese medicinals. The Chinese medicinals were used for 1 480 times in total and the top 20 included Paeoniae Radix Rubra,Angelicae Sinensis Radix,Salviae Miltiorrhizae Radix et Rhizoma,Chuanxiong Rhizoma and Liquidambaris Fructus,etc. The Chinese medicinals were mainly warm, cold and neutral in nature, and bitter, sweet and acrid in flavor, and their main meridian entries were the liver, kidney and spleen. The association rules analysis suggested the medicinal combinations of Spargani Rhizoma-Curcumae Rhizoma and Carthami Flos-Angelicae Sinensis Radix, etc. The cluster analysis presented six new medicinal combinations, and the Class 1 medicinal combinations (Curcumae Rhizoma, Spargani Rhizoma, Liquidambaris Fructus,Gleditsiae Spina and Salviae Miltiorrhizae Radix et Rhizoma) could be the core prescription for treating TI by Chinese medicinals. The β-sitosterol,quercetin,luteolin,cryptotanshinone and fisetin were the critical ingredients of core prescription for TI and these ingredients may have an effect on the targets of tumor necrosis factor (TNF),tumor protein 53 (TP53), interleukin (IL) 6, estrogen receptor 1 (ESR1) and B-cell lymphoma-2 (BCL2) by signaling pathways in cancer, hypoxia inducible factor-1 (HIF-1), mitogen-activated protein kinase (MAPK) and FoxO. Conclusion:The Chinese medicinals for promoting qi and activating blood,and resolving stasis and relieving pain are mainly used for TI in clinical, and the core prescription may exert its curative effect on TI by playing a role in cells proliferation, cellular differentiation and inflammatory responses through signaling pathways in cancer, HIF-1, MAPK and FoxO.
