Abstract: Objective: To observe the wound repair ability and mechanism of action of the extract of Melastoma dodecandrum Lour. on model rats with superficial Ⅱdegree burn. Methods: A total of 50 rats were randomly divided into the high-dose group of extract of Melastoma Dodecandrum Lour., the lowdose group of extract of Melastoma dodecandrum Lour., the Kangfuxin Liquid group, and the model control group,with normal rats as the blank control group. Except for the blank control group,burn models were prepared using the steam burn method in all the other groups. The Kangfuxin Liquid group and the groups of extract of Melastoma dodecandrum Lour. were given administration daily at a fixed time and treated continuously for 21 days. During the treatment period, the wound-healing status of rats in each group was recorded. After 21 days of treatment,ELISA method was used to detect the expression levels of superoxide dismutase (SOD),malondialdehyde (MDA),interleukin-6 (IL-6),interleukin-10 (IL-10),and tumor necrosis factor- α (TNF- α) in serum in each group. HE staining was used to observe pathological changes such as changes in skin tissue structure,infiltration of inflammatory cells,and neovascularization in the burn area. Results:On the seventh,fourteenth,and twenty-first days after burns,the wound area in the low-dose group of extract of Melastoma dodecandrum Lour. was significantly larger than that in the Kangfuxin Liquid group,and the healing rate was lower than that in the Kangfuxin Liquid group (P<0.05). On the seventh, fourteenth, and twenty-first days after burns, the healing status of wound area and healing rate in the high-dose group of extract of Melastoma dodecandrum Lour. were comparable to those in the Kangfuxin Liquid group (P>0.05). The levels of SOD and IL-10 in serum in the model control group were decreased when compared with those in the blank control group (P<0.05),and the levels of MDA, IL-6, and TNF- α were increased when compared with those in the blank control group (P<0.05). Compared with those in the model control group,the levels of SOD and IL-10 in serum in the Kangfuxin Liquid group,the high-dose group of extract of Melastoma dodecandrum Lour.,and the low-dose group of extract of Melastoma dodecandrum Lour. were increased (P<0.05),and the levels of MDA,IL-6,and TNF- α were decreased (P<0.05). Conclusion: The extract of Melastoma dodecandrum Lour. can inhibit the inflammatory and oxidative stress responses caused by burns, increase the number of neovascularization in wound tissue,and promote wound repair in rats with superficial II degree burns.
