    目的:观察熊果酸对雌激素受体ERα、ERβ表达的影响,研究熊果酸抑制甲状腺癌BCPAP细胞增 殖、迁徙、侵袭的机制。方法:Western blot 实验检测人正常甲状腺细胞(Nthy-ori3-1 细胞) 与甲状腺癌 BCPAP细胞中ERα、ERβ的蛋白表达,CCK-8实验检测熊果酸对甲状腺癌BCPAP细胞增殖的影响,细胞划痕 实验、Transwell实验检测熊果酸对甲状腺癌BCPAP细胞迁徙、侵袭能力的影响。Western blot实验检测熊果酸 对甲状腺癌BCPAP细胞雌激素受体ERα、ERβ表达的影响。结果:Nthy-ori3-1细胞与BCPAP细胞比较,ERα 的表达水平较低(P<0.05),ERβ的表达水平较高(P<0.05)。不同浓度的熊果酸(4、8、16、32、64 μmol/L) 对BCPAP 细胞增殖具有抑制作用,呈时间和浓度依赖性(P<0.05)。同时间点,B4 组(4 μmol/L)、B8 组(8 μmol/L) BCPAP细胞迁移力低于B0组(0 μmol/L)(P<0.05)。与B0组比较,B4组、B8组BCPAP细胞 侵袭数明显降低(P<0.05)。熊果酸作用24 h后,B4组、B8组ERα表达水平低于B0组(P<0.05),ERβ表达 水平高于B0组(P<0.05)。结论:熊果酸可能通过雌激素受体ERα、ERβ降低甲状腺癌BCPAP细胞增殖、迁 徙、侵袭的能力。

    Abstract:

    Abstract: Objective: To observe the effect of ursolic acid on the expressions of estrogen receptors ERα and ERβ,and to study the mechanism of ursolic acid in inhibiting proliferation,migration and invasion of thyroid cancer BCPAP cells. Methods:The protein expressions of ERα and ERβ in human normal thyroid cells (Nthy-ori3-1 cells) and thyroid cancer BCPAP cells were detected by Western blot. The effect of ursolic acid on the proliferation of thyroid cancer BCPAP cells was detected by CCK-8 assay. The effect of ursolic acid on the migration and invasion of thyroid cancer BCPAP cells was detected by cell scratch assay and Transwell assay. The effect of ursolic acid on the expressions of estrogen receptor ERα and ERβ in thyroid cancer BCPAP cells was detected by Western blot. Results: Compared with BCPAP cells, the expression level of ERα was lower in Nthy-ori3-1 cells (P<0.05), and the expression level of ERβ was relatively high (P<0.05). Ursolic acid at different concentrations (4, 8, 16, 32, 64 μmol/L) inhibited the proliferation of BCPAP cells in a time- and concentration-dependent manner (P<0.05). The migration of BCPAP cells in the B4 group (4 μmol/L) and the B8 group (8 μmol/L) was lower than that in the B0 group (0 μmol/L) at the same time point (P<0.05). Compared with the B0 group, the number of invasive BCPAP cells in the B4 and B8 groups was significantly decreased (P<0.05). After 24 hours of treatment with ursolic acid,the expression level of ERα in the B4 and B8 groups was lower than that in the B0 group (P<0.05), and the expression level of ERβ was higher than that in the B0 group (P<0.05). Conclusion: Ursolic acid may reduce the proliferation, migration and invasion of thyroid cancer BCPAP cells through estrogen receptors ERα and ERβ.
