Abstract: Objective: To investigate the signal transduction mechanism of electroacupuncture in spontaneously hypertensive left ventricular hypertrophy rats. Methods: Spontaneous hypertensive rats (SHR) were randomly divided into hypertensive left ventricular hypertrophy group,electroacupuncture point group,electroacupuncture non- point group,AT1- R siRNA group and STAT siRNA group. Wistar- Kyoto rats with normal blood pressure were used as control group,with 20 rats in each group. Blood pressure, cardiac structure changes,myocardial fibrosis,angiotensin Ⅱ(AngⅡ) concentration,angiotensin receptor 1 (AT1) mRNA,Janus kinase and expression of transcriptional signal transducers and activators (JAK/STAT) protein were observed before and after treatment. Results: The systolic blood pressure of rats in the electroacupuncture point group decreased after treatment,but the systolic blood pressure of rats in the left ventricular hypertrophy group,the non- point group of electroacupuncture,the AT1- R siRNA group and the STAT siRNA group did not decrease significantly after treatment,and the differences were statistically significant (P<0.05). The left ventricular mass index (LVMI) in electroacupuncture point group was lower than that in hypertensive left ventricular hypertrophy group,electroacupuncture non-point group,AT1-R siRNA group and STAT siRNA group, and the difference was statistically significant (P<0.05). The left ventricular short axis shortening rate (LVFS) in electroacupuncture point group was higher than that in hypertensive left ventricular hypertrophy group,electroacupuncture non-point group,AT1-R siRNA group and STAT siRNA group,and the difference was statistically significant (P<0.05). HE staining showed that the heart volume and cross- sectional area of myocardium of rats in the electroacupuncture point group were smaller than those in the hypertensive left ventricular hypertrophy group. The levels of Ang Ⅱ in myocardial tissue,serum procollagen (PCⅢ),laminin (LN) and hyaluronic acid (HA) in electroacupuncture point group were lower than those in hypertensive left ventricular hypertrophy group, with statistical significance (P<0.05). The AT1mRNA level of myocardial tissue in electroacupuncture point group was lower than that in hypertensive left ventricular hypertrophy group, and the difference was statistically significant (P<0.05). The expressions of JAK1 and STAT3 in hypertensive left ventricular hypertrophy group,electroacupuncture non-point group and AT1-R siRNA group were high,while the expressions of JAK1 and STAT3 in electroacupuncture point group were decreased,with statistical significance (P<0.05). Conclusion: Electroacupuncture may reverse left ventricular hypertrophy by inhibiting the expressions of related cytokines and proteins in JAK/STAT pathway mediated by AT1.
