Abstract:Objective:To optimize the extraction process of Liandou Qingmai Mixture. Methods:The contents of forsythiin and berberine in Liandou Qingmai Mixture were determined by high performance liquid chromatography (HPLC). The transfer rate of forsythiin and berberine and extract yield were used as comprehensive evaluation indexes. The optimal extraction process was determined by L9(34) orthogonal test. The effects on the contents of forsythiin and berberine were investigated by atmospheric concentration and vacuum concentration. Results: Forsythiin and berberine had a good linear relationship in 5.516- 110.316 μg/mL and 4.292- 85.833 μg/mL, respectively. The average recoveries were 101.31% and 101.97%,RSD were 1.58% and 1.76% (n=6). The optimal extraction process was to soak Liandou Qingmai Mixture with 10 times water for 30 minutes and extract twice for 1.5 hours each time, and different concentration methods had no significant effect on the contents of forsythiin and berberine. Conclusion: The optimized extraction technology is reasonable and feasible, which provides scientific basis for the large-scale production and research and development of Liandou Qingmai Mixture.
