Abstract: Objective: To investigate the effects of Compound Qishao Jiangya Tablets on blood pressure and left ventricular hypertrophy in hypertensive rats via miR- 214. Methods:The blood pressure changes of rats were monitored in real time by using a non-invasive tail artery blood pressure measurement system.The structure of rat heart was measured by echocardiography.Cardiac gross morphology, cardiac index and left ventricular mass index were observed.Hematoxylin eosin staining (HE) was used to detect the pathological changes of myocardium in rats. Radioimmunoassay and enzyme-linked immunosorbent assay (ELISA) were used to detect the serum indexes of ventricular hypertrophy and the expression of angiotensin Ⅱ (Ang Ⅱ) in myocardium and plasma of rats. RT- PCR was used to detect the changes of miR- 214 in hypertrophic cardiomyocytes after the intervention of Compound Qishao Jiangya Tablets. Results: The results of blood pressure showed that Compound Qishao Jiangya Tablets could improve the blood pressure of hypertensive rats. The results of echocardiography showed that Compound Qishao Jiangya Tablets could improve the cardiac function of hypertensive rats. The pathological results showed that Compound Qishao Jiangya Tablets could improve the ventricular hypertrophy in hypertensive rats. The results of radioimmunoassay and ELISA showed that Compound Qishao Jiangya Tablets could reduce the expression of serum type procollagen (PC Ⅲ), laminin (LN), hyaluronic acid (HA) and Ang Ⅱ in myocardium and plasma of rats. The RT-PCR results showed that Compound Qishao Jiangya Tablets inhibited the expression of miR- 214 in hypertrophic cardiomyocytes, and the difference of the above results was statistically significant (P<0.05). Conclusion: Compound Qishao Jiangya Tablets may improve blood pressure and ventricular hypertrophy in hypertensive rats by inhibiting the expression of miR-214.
