    摘要:

    目的:基于网络药理学联合GEO 芯片差异基因分析方法探讨肾气丸治疗肾小管上皮间质转化(EMT) 的作用机制,动物实验进一步验证肾气丸治疗EMT 的作用靶点。方法:依据中药系统药理学数据库与分析平台(TCMSP) 和BATMAN-TCM 数据库筛选出肾气丸的活性成分及其对应靶点,利用GEO 数据库获取疾病相关靶点, 通过Cytoscape3.7.0 软件绘制“ 药物-成分-靶点” 网络。使用STRING 数据库与Cytoscape3.7.0 软件绘制核心靶点蛋白相互作用网络,并利用R 语言对核心靶点进行GO 功能和KEGG 通路富集分析。通过实时荧光定量PCR 验证肾气丸对苏氨酸蛋白激酶1(AKT1)、细胞分裂周期蛋白20(CDC20)、表皮生长因子受体(EGFR)、核受体辅激活蛋白1(NCOA1)、细胞周期蛋白B1(CCNB1) mRNA 的影响。结果:共得到肾气丸活性成分229 个,预测作用靶点共1 331 个,与EMT GEO 芯片共同靶点共221 个。肾气丸治疗EMT 主要作用于AKT1、CDC20、EGFR、NCOA1、CCNB1 等靶点。GO 富集分析涉及到营养物质反应、金属离子反应、氧化应激反应等多种生物过程,KEGG 富集分析得到cAMP 信号通路、钙信号通路、甲状腺激素信号通路、催产素信号通路、HIF-1 信号通路等多条信号通路。实时荧光定量PCR 证明肾气丸治疗可升高腺嘌呤造模小鼠肾组织中AKT1 mRNA 表达,降低CDC20、EGFR 和CCNB1 mRNA 表达(P<0.05)。结论:基于网络药理学方法及实验验证,初步阐明肾气丸可通过调节AKT1、CDC20、EGFR、CCNB1 mRNA 表达来改善EMT,其作用机制可能与cAMP 信号通路、钙信号通路等多条信号通路有关。

    Abstract:

    Abstract: Objective: To discuss the mechanism of Shenqi pills in the treatment of epithelialmesenchymal transformation(EMT) of renal tubules on the basis of the method of network pharmacology combined with differential gene analysis by GEO chip,and to further verify the targets of Shenqi Pill in the treatment of EMT of renal tubules by animal experiments. Methods: According to Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP) and BATMAN- TCM database, the active components and their corresponding targets of Shenqi pills were screened. The disease-related targets were obtained by GEO database and “drug- component- target” network was drawn by Cytoscape3 7.0 software. The interaction network of core target proteins was drawn by using STRING database and Cytoscape3 7.0 software,and R language was used to annotate the GO function and perform the pathway enrichment analysis of KEGG. The effects of Shenqi pills on threonine protein kinase 1(AKT1), cell division cycle 20(CDC20), epidermal growth factor receptor(EGFR), nuclear receptor coactivator 1 (NCOA1) and cyclin B1(CCNB1) mRNA were verified by real- time fluorescence quantitative PCR. Results: A total of 229 active components of Shenqi pills were obtained, including 1 331 predicted targets, and 221 common targets with EMT of renal tubules on GEO chip. Shenqi pills mainly acted on AKT1,CDC20, EGFR,NCOA1,CCNB1 and other targets in the treatment of EMT of renal tubules. Go enrichment analysis involved many biological processes such as nutrient reactions, metal ion reactions and oxidative stress reactions. KEGG enrichment analysis obtained many signal pathways such as cAMP signal pathway, calcium signal pathway, thyroid hormone signal pathway, oxytocin signal pathway and HIF- 1 signal pathway. Real- time fluorescence quantitative PCR showed that Shenqi pills could increase the mRNA expression of AKT1 and reduce the mRNA expression of CDC20,EGFR and CCNB1 in the kidney tissues of adenine modle mice(P<0.05). Conclusion: Based on the network pharmacology and experimental verification,it is preliminarily clarified that Shenqi pills can improve EMT of renal tubules by regulating the mRNA expression of AKT1, CDC20, EGFR and CCNB1, and its mechanism may be related to multiple signal pathways such as cAMP signal pathway and calcium signal pathway.
