    摘要:

    目的:探讨补肾解毒方对放射致感染性肺损伤的防护作用。方法:选取72 只C57BL/10J 小鼠,随机分为空白对照组、单纯照射组、氨磷汀组、中药组,每组18 只,建立各组小鼠模型,在实验开始后第11 天,空白对照组小鼠不接受照射,余各组小鼠均接受一次性6 Gy 60Coγ 射线全身照射,其中氨磷汀组在照射前30 min 内注射氨磷汀。并于照射后第1 天、第7 天、第14 天每组随机抽取6 只小鼠进行取材,计算肺脏系数,采用酶联免疫吸附实验(ELISA) 检测降钙素原(PCT)、C-反应蛋白(CRP)、白细胞介素-6(IL-6),并在光学显微镜下观察小鼠肺组织病理形态学变化。结果:照射后第1 天,空白对照组肺系数水平低于余3 组(P<0.05),同时单纯照射组高于氨磷汀组和中药组(P<0.05);照射后第7 天,空白对照组肺系数水平低于单纯照射组和氨磷汀组(P<0.05),同时单纯照射组高于氨磷汀组和中药组(P<0.05);照射后第14 天,单纯照射组肺系数水平高于余3 组(P<0.05)。单纯照射组病理形态学变化最明显,氨磷汀组和中药组可明显减轻肺组织病理变化,其中中药组优于氨磷汀组。照射后第1 天,空白对照组PCT、CRP、IL-6 水平显著低于余3 组(P<0.05),同时单纯照射组CRP、IL-6 水平高于氨磷汀组与中药组(P<0.05);照射后第7 天,PCT、CRP、IL-6 水平中,空白对照组低于余3 组(P<0.05),单纯照射组高于氨磷汀组和中药组(P<0.05),同时中药组低于氨磷汀组(P<0.05);照射后第14 天,空白对照组PCT、IL-6 水平低于单纯照射组和氨磷汀组(P<0.05),单纯照射组高于氨磷汀组和中药组(P<0.05),中药组低于氨磷汀组(P<0.05),单纯照射组CRP 水平高于余3 组(P<0.05)。结论:补肾解毒方能够减轻放射致感染性肺损伤的感染程度,降低PCT、CRP 水平,发挥抗炎抗损伤作用。

    Abstract:

    Abstract: Objective: To explore the protective effect of Bushen Jiedu decoction on radiation- induced infectious lung injury. Methods:72 C57BL/10J mice were randomly divided into blank control group,simple irradiation group,amifostine group and traditional Chinese medicine group, with 18 mice in each group. The mouse models of each group were established. On the 11th day after the beginning of the experiment,the mice in blank control group did not receive irradiation, and the other mice in each group received one-time 6 Gy 60Co γ 30 min before the whole body irradiation with fostine. Six mice in each group were randomly selected on the 1st , 7th and 14th day after irradiation. The lung coefficient was calculated. Procalcitonin(PCT), C- reactive protein(CRP) and interleukin- 6(IL- 6) were detected by enzyme- linked immunosorbent assay(ELISA),and the pathomorphological changes of mouse lung were observed under optical microscope. Results: On the first day after irradiation, the lung coefficient of the blank control group was lower than that of the other three groups(P<0.05), and that of the simple irradiation group was higher than that of amifostine group and traditional Chinese medicine group(P<0.05). On the 7th day after irradiation, the lung coefficient level in the blank control group was lower than that in the simple irradiation group and amifostine group(P<0.05),while that in the simple irradiation group was higher than that in the amifostine group and traditional Chinese medicine group(P<0.05). On the 14th day after irradiation,the lung coefficient in the simple irradiation group was higher than that in the other three groups(P<0.05).The pathomorphological changes were the most obvious in the simple irradiation group. Amifostine group and traditional Chinese medicine group could significantly reduce the pathological changes of lung tissue, and the traditional Chinese medicine group was better than amifostine group. On the first day after irradiation, the levels of PCT, CRP and IL- 6 in the blank control group were significantly lower than those in the other three groups(P<0.05),while the levels of CRP and IL- 6 in the simple irradiation group were higher than those in amifostine group and traditional Chinese medicine group(P<0.05). On the 7th day after irradiation,the levels of PCT,CRP and IL-6 in the blank control group were lower than those in the other three groups(P< 0.05), those in the simple irradiation group were higher than those in the amifostine group and the traditional Chinese medicine group(P<0.05),and those in the traditional Chinese medicine group were lower than those in the amifostine group (P<0.05). On the 14th day after irradiation,the levels of PCT and IL-6 in the blank control group were lower than those in the simple irradiation group and amifostine group(P<0.05), higher than that in the amifostine group and traditional Chinese medicine group(P<0.05),lower than that in the amifostine group(P<0.05),and the level of CRP in the simple irradiation group was higher than that in the other three groups(P<0.05). Conclusion:Bushen Jiedu decoction can reduce the infection degree of radiation-induced lung injury,reduce the levels of PCT and CRP,and play an anti-inflammatory and anti-injury role.
