    目的:研究民族药傣百解抗脓毒症的作用,揭示其抗脓毒症免疫学机制。方法:采用细菌脂多糖腹腔注射构建脓毒症模型,随机分为正常组、模型组和傣百解醇提物100 mg/kg 和傣百解醇提物200 mg/kg 剂量组,药物干预24 h 和48 h 后,统计生存率;HE 染色检测肺和肝脏组织病理损伤程度;流式细胞术检测淋巴细胞亚群(CD3+ T 细胞、CD4+ T 细胞、CD8+ T 细胞、TCR+ T 细胞和B 细胞) 比例;流式细胞术测定T 细胞亚群(Th1、Th17 和Treg) 表达。结果:与脓毒症模型组比较,傣百解醇提物提高了脓毒症模型小鼠生存率,减少炎症细胞向肺和肝组织浸润,对肺和肝病理损伤具有较好的保护效应;傣百解醇提物药物干预可下调B220+ B 淋巴细胞表达(P<0.05),然而对脓毒症模型小鼠CD3+ T 细胞、CD4+ T 细胞、CD8+ T 细胞和TCR+ T 细胞的比例无明显的影响(P>0.05);可显著上调CD4+ Foxp3+ Treg 细胞的比例(P<0.05),然而对Th1 细胞(CD4+ IFN-+T 细胞) 和Th17细胞(CD4+ IL-17A+T 细胞) 表达无影响(P>0.05)。结论:傣药傣百解对抗脓毒症具有显著的治疗作用,其机制可能与诱导调节性T 细胞介导免疫自稳和耐受作用有关。

    Abstract:

    Abstract:Objective:To study the anti-sepsis effect of Marsdenia Tenacissima(Roxb) Moon(DBJ),a national medicine, and to reveal its anti sepsis immunological mechanism. Methods: The sepsis model was established by intraperitoneal injection of bacterial lipopolysaccharide. It was randomly divided into normal control group, model group and DBJ alcohol extract 100 mg/kg and DBJ alcohol extract 200 mg/kg groups. The survival rate was counted after 24 and 48 hours of drug intervention.HE staining was used to detect the degree of pathological injury of lung and liver. The proportion of lymphocyte subsets(CD3+ T cells,CD4+ T cells,CD8+ T cells,TCR+ T cells and B cells) was detected by flow cytometry.The expression of T cell subsets(Th1,Th17 and Treg) was measured by flow cytometry. Results:Compared with sepsis model group,DBJ alcohol extract improved the survival rate of sepsis model mice,reduced the infiltration of inflammatory cells into lung and liver tissue, and had a better protective effect on lung and liver pathological injury. DBJ alcohol extract decreased the expression of B220+ B lymphocytes(P<0.05),but had no significant effect on the proportion of CD3+ T cells,CD4+ T cells, CD8 + T cells and TCR + T cells in sepsis model mice(P>0.05),and significantly increased the proportion of CD4 + Foxp3 + Treg cells(P<0.05),but had no effect on the expression of Th1 cells(CD4+ IFN-+ T cells) and Th17 cells(CD4+ IL-17A+ T cells)(P> 0.05). Conclusion:DBJ has a significant therapeutic effect on sepsis,and its mechanism may be related to the induction of regulatory T cells and the mediation of immune homeostasis and tolerance.
