    目的:探讨枳术汤对急性脑梗死(ACI) 大鼠神经功能及海马组织神经轴突生长抑制因子-A(Nogo-A) 及溶血磷脂酸(LPA)蛋白表达的影响。方法:采用线栓法制备急性脑梗死大鼠模型,将造模成功的大鼠随机分成模型组、枳术汤低(6.9 g/kg)、中(13.8 g/kg)、高(27.6 g/kg) 剂量组,每组14 只。取14 只大鼠(只分离血管,不阻塞血流) 作为假手术组,枳术汤低、中、高剂量组分别灌胃6.9 g/kg、13.8 g/kg 和27.6 g/kg 的枳术汤,假手术组和模型组灌胃给予等体积0.9%氯化钠溶液。连续给药14 d后,测定大鼠神经功能缺损评分、血清乳酸脱氢酶(LDH) 和谷胱甘肽过氧化物酶(GSH-Px) 的含量、脑梗死体积比例、脑含水量和海马组织中Nogo-A 和LPA 蛋白水平。结果:与假手术组相比,模型组大鼠神经功能缺损评分、脑含水量、脑梗死体积比例、血清LDH、海马组织Nogo-A 和LPA 蛋白水平显著增加,血清GSH-Px 含量显著降低(P<0.05);与模型组相比,枳术汤低、中、高剂量组大鼠神经功能缺损评分、脑含水量、脑梗死体积比例、血清LDH、海马组织Nogo-A 和LPA 蛋白水平降低,血清GSH-Px 含量增加,呈剂量依赖性(P<0.05)。结论:枳术汤能改善ACI 大鼠神经功能,减少脑梗死体积比例,其机制可能与枳术汤抑制海马组织Nogo-A 和LPA 蛋白表达有关。

    Abstract:

    Abstract:Objective:To investigate the effects of Zhizhu tang on neural function and the expression of neurite outgrowth inhibitor-A(Nogo-A) and lysophosphatidic acid(LPA) in hippocampus of rats with acute cerebral infarction(ACI). Methods:The rat model of ACI was prepared by thread thrombus method. The successful rats were randomly divided into model group,low (6.9 g/kg), medium(13.8 g/kg) and high(27.6 g/kg) dose groups of Zhizhu tang, with 14 rats in each group. 14 rats (only isolated blood vessels without blocking blood flow) were taken as the sham operation group. The low,medium and high dose groups of Zhizhu tang were gavaged with 6.9 g/kg,13.8 g/kg and 27.6 g/kg of Zhizhu tang respectively. The sham operation group and model group were gavaged with equal volume of 0.9% sodium chloride solution. After continuous administration for 14 days,the neurological deficit score,the content of serum lactate dehydrogenase(LDH) and glutathione peroxidase(GSHPx), the volume proportion of cerebral infarction, the brain water content and Nogo- A and LPA protein levels in hippocampus. Results: Compared with the sham operation group, the neurological deficit score, brain water content, cerebral infarction volume ratio,serum LDH,Nogo-A and LPA protein levels in hippocampus were significantly increased, and the content of serum GSH-Px was significantly decreased in the model group(P<0.05).Compared with the model group, the neurological deficit score,brain water content,cerebral infarction volume ratio,serum LDH,Nogo-A and LPA protein levels in hippocampus decreased, and the content of serum GSH- Px increased in a dose- dependent manner(P<0.05). Conclusion:Zhizhu tang can improve neurological function and reduce the volume proportion of cerebral infarction in ACI rats. Its mechanism may be related to Zhizhu tang inhibiting the expression of Nogo-A and LPA protein in hippocampus.
