Abstract:Objective:To investigate the action mechanism of Tongguan capsules for atherosclerotic diseases by means of network pharmacology. Methods:Pharmaceutical compounds and targets of Tongguan capsules were screened by TCMSP database and literature mining; the related targets of atherosclerotic diseases were retrieved through GeneCards and DisGeNET databases; a Tongguan capsules- target- disease network and a core network were constructed through Cytoscape 3.8.0 software and its PPI network was constructed through STRING database. Potential targets of Tongguan capsules for atherosclerotic diseases were selected for KEGG and GO enrichment analysis on DAVID website. R software was applied to visualize the above signal pathway, biological process, molecular function and cell composition information. Results: A total of 123 compounds, 272 drug targets and 111 corresponding compounds of Tongguan capsules were obtained; there were 576 targets of atherosclerotic diseases. The intersection graph of drug targets of Tongguan capsules and targets of atherosclerosis was formed,with a total of 61 common targets such as NR3C1,DPP4,ESR1,MAPK14, PPARG and PTGS2,and 101 corresponding compounds. There were 56 interconnected nodal points and 288 edges in PPI network. After KEGG and GO analysis, 22 pathways for Tongguan capsules to act on atherosclerosis were obtained, including NOD- like receptor signaling pathway, cancer pathway and VEGF signaling pathway. Conclusion: Tongguan capsules can regulate insulin signaling pathway,play a role in inhibiting inflammatory responses,regulating metabolism and anti- oxidative stress as well as intervene in the occurrence and development of atherosclerosis by acting on 61 common targets such as NR3C1,DPP4,ESR1,MAPK14,PPARG and PTGS2.
