Xiaoyao Powder Has Effect on Expression of HSPA1A Gene in Hippocampus Tissue of Rats with Chronic Stress

目的:探讨并比较中药复方逍遥散和抗抑郁药百忧解(盐酸氟西汀) 对慢性应激大鼠海马组织中HSPA1A 基因表达的干预作用。方法:将SPF 级Wistar 大鼠120 只随机分为空白组、模型组、逍遥散组、百忧解组,除空白组外,采用足底电击、冰水游泳、禁食24 h、禁水24 h、束缚4 h 等应激源对大鼠进行慢性应激刺激。空白组和模型组每只大鼠按照10 mL/ (kg · d) 的剂量灌胃生理盐水,百忧解组和逍遥散组每只大鼠分别按10 mL/(kg · d) 的剂量灌胃百忧解和逍遥散。评价模型制备21 d、28 d、35 d 的造模效果;荧光定量PCR 检测实验21 d、28 d、35 d 时大鼠海马组织中HSPA1A mRNA 的表达。结果:与空白组比较,模型组大鼠造模21 d、28 d、35 d,体质量均明显减轻(P<0.01),水平运动得分及垂直运动得分均降低(P<0.05,P<0.01),血清皮质酮含量升高(P<0.05,P<0.01),海马组织中HSPA1A mRNA 相对表达量降低(P<0.01)。与模型组比较,逍遥散组和百忧解组大鼠造模21 d 体质量均升高(P<0.05,P<0.01);逍遥散组大鼠造模21 d、28 d、35 d 水平运动得分升高(P<0.05,P<0.01);逍遥散组和百忧解组大鼠造模21 d、35 d 垂直运动得分均升高(P<0.05);逍遥散组及百忧解组大鼠造模21 d、28 d、35 d 血清皮质酮含量降低(P<0.05,P<0.01);逍遥散组大鼠造模21 d 海马组织中HSPA1AmRNA 相对表达量升高(P<0.01);百忧解组大鼠造模35 d 海马组织中HSPA1A mRNA 相对表达量升高。与百忧解组比较,逍遥散组大鼠造模35 d 水平运动得分较高(P<0.01),HSPA1A mRNA 相对表达量较低(P<0.01)。结论:逍遥散对慢性应激造成的HSPA1A mRNA 表达降低的调节作用随时间延长逐渐减弱,百忧解对慢性应激引起的HSPA1A mRNA 表达下调的干预作用则在实验35 d 时才突显,说明中药复方的治疗作用具有时效性,应该随证变化及时调整方药;抗抑郁药起效慢,临床治疗慢性应激相关病证考虑中西医结合治疗,可能会取得更理想的疗效。
Abstract:
Abstract: Objective: To discuss the effect of compound Xiaoyao powder of Chinese medicine and antidepressant, namely fluoxetine(fluoxetine hydrochloride) on intervening expression of HSPA1A gene in hippocampus tissue of rats with chronic stress respectively, and compare the intervention effect of the two applications above. Methods: A total of 120 Wistar rats of SPF level were randomly divided into the blank group,the model group,the Xiaoyao powder group,and the fluoxetine group. Except for the blank group, in the other groups, chronic stress stimulation was imposed upon rats by applying electric foot-shock,swimming in ice water,fasting for one day,water deprivation for one day,restraint for four hours,and other stressors. The blank group and the model group were given gastric perfusion of normal saline at a dose of 10 mL/(kg · d). The fluoxetine group and the Xiaoyao powder group were given gastric perfusion of fluoxetine and Xiaoyao powder respectively at a dose of 10 mL/(kg·d). On the 21st,28th,and 35th day of model preparation,the modeling effect was evaluated. On the 21st,28th,and 35th day of experiment,the expression of mRNA of HSPA1A in hippocampus tissue of rats were detected by fluorescent quantitative PCR. Results:On the 21st,28th,and 35th day of modelling,when compared with those in the black group,body mass in the model group was significantly reduced(P<0.01);the scores of horizontal motion and vertical motion were decreased(P<0.05,P<0.01);the corticosterone content in serum was increased(P<0.05, P<0.01); the relative expression quantity of mRNA of HSPA1A in hippocampus tissue was decreased(P<0.01). When compared with those in the model group,body mass in the Xiaoyao powder group and the fluoxetine group was increased on the 21st day of modelling(P<0.05,P<0.01);the scores of horizontal motion in the Xiaoyao powder group were increased on the 21st,28th,and 35th day of modelling(P<0.05,P<0.01);the scores of vertical motion in the Xiaoyao powder group and the fluoxetine group were increased on the 21st and 35th day of modelling(P<0.05);the corticosterone content in serum in the Xiaoyao powder group and the fluoxetine group was decreased on the 21st, 28th, and 35th day of modelling(P<0.05, P< 0.01); the relative expression quantity of mRNA of HSPA1A in hippocampus tissue was increased in the Xiaoyao powder group on the 21st day of modelling(P<0.01),as well as in the fluoxetine group on the 35th day of modelling. On the 35th day of modelling, when compared with that in the fluoxetine group, the score of horizontal motion in the Xiaoyao powder group was higher(P<0.01), and the relative expression quantity of mRNA of HSPA1A was lower(P<0.01). Conclusion: Xiaoyao powder had regulatory effect on decrease of expression of mRNA of HSPA1A induced by chronic stress,which was gradually weakened as time went by. Fluoxetine had intervention effect on down-regulation of mRNA expression of HSPA1A induced by chronic stress;it was on the 35th day of experiment that the intervention effect became significant. From the two findings above,it is illustrated that the curative effect of compound prescription of Chinese medicine has chronergy,which means that prescriptions and medicine should be regulated in time with changes in syndromes. Based on the fact that antidepressants take effect more slowly,integrated Chinese and western medicine therapy is taken into consideration in the clinical treatment of diseases and syndromes related to chronic stress,which probably obtains more ideal curative effect.
