Abstract:Objective:To study the effect of Radix Alangii Chinensis on metabolism of plasma and urine in rats based on gas chromatography-mass spectrometry(GC-MS) technology. Methods:A total of 32 SD rats were divided into the control group and the Radix Alangii Chinensis groups with low,medium and high dose respectively,8 rats in each group. The control group was given normal saline by gavage,and the Radix Alangii Chinensis groups with low,medium and high dose were given Radix Alangii Chinensis decocted fluid by gavage at the doses of 0.25, 0.50 and 1.00 g/(kg · d) respectively. All the groups were treated for 14 days. On the 15th day,samples of plasma and urine in each group were collected for derivation, and then GC-MS analysis was performed. Results:The separation effect of partial least squares regression analysis(PLS-DA) scores among the groups was not good,and there was intersection among the groups,but the separation effect of plasma was better than that of urine samples. The partial least squares regression 3D model(PLS- 3D) scores in the Radix Alangii Chinensis groups with low,medium and high dose respectively could be distinguished from that in the control group;the PLS-3D scores were better than PLS-DA scores. Compared with those in the control group respectively,the urea content in plasma in the Radix Alangii Chinensis group with high dose was increased(P<0.05),and the content of urinary glutaric acid was decreased(P<0.05);the contents of glucose,flavianic acid and succinamic acid in the Radix Alangii Chinensis groups with medium and high dose respectively were decreased(P<0.05); the content of xylitol in urine in the Radix Alangii Chinensis group with low dose was increased(P < 0.05). Conclusion : The study of metabonomics based on GC- MS technology finds that Radix Alangii Chinensis can induce the metabolic disorder of plasma and urine in rats, and provide references on toxicology and pharmacology for clinical safe medication.
