乌药叶提取物对高脂血症大鼠肝脏组织病理学及TLR-4/NF-κB 信号通路的影响

    摘要:

    目的:探讨乌药叶提取物对高脂血症模型大鼠肝脏组织病理学及Toll 样受体4(TLR-4)/核转录因子-κB(NF-κB) 信号通路的影响。方法:将40 只SPF 级雄性大鼠随机分为对照组10 只和造模组30 只。造模组通过高脂饮食建立高脂血症大鼠模型,模型制备成功后,将造模组大鼠随机分为模型组与乌药叶高、低剂量组。乌药叶高、低剂量组分别按1.6 g/kg、0.8 g/kg 的剂量灌胃乌药叶提取物,模型组及对照组给予等容积的蒸馏水。连续给药8 周后,HE 染色后观察大鼠肝脏组织病理学变化,Western blot 法检测其肝脏TLR-4、NF-κB、肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2) 蛋白表达。结果:与对照组比较,模型组大鼠肝细胞排列紊乱、炎性细胞浸润、并呈现大小不一的脂滴,TLR-4、NF-κB、TNF-α、IL-2 蛋白表达均增加(P<0.05);与模型组比较,高、低剂量组大鼠肝脏病理形态接近对照组,肝细胞排列较为整齐,炎性细胞浸润明显减轻,肝细胞内小脂滴数量减少,细胞空泡化明显改善,TLR-4、NF-κB、TNF-α、IL-2 蛋白表达均降低(P<0.05),其中以乌药叶高剂量组效果更显著。结论:乌药叶提取物对高脂血症模型大鼠的肝组织病理改变有明显的改善作用,其作用机制可能是通过阻断TLR-4/NF-κB 信号通路,减少TNF-α、IL-2 蛋白表达实现的。

    Abstract:

    Abstract:Objective:To discuss the effect of extract of linderae aggregate leaf on pathology of the liver tissues and Toll- like receptor 4(TLR-4)/nuclear factor-κB(NF-κB) signaling pathway in the rat models with hyperlipidemia. Methods:A total of 40 cases of male rats of SPF level were randomly divided into the control group and the modeling group,10 and 30 rats in each group respectively. The rat models with hyperlipidemia were established through high- fat diet in the modeling group. After successful establishment,the rats in the modeling group were randomly divided into the model group and the linderae aggregate groups of high-dose and low-dose. The linderae aggregate groups of high-dose and low-dose were administered with gastric perfusion with extract of linderae aggregate leaf in 1.6 g/kg and 0.8 g/kg respectively,and the model group and the control group were given distilled water of same volume. After eight weeks of continuous medication, the changes in pathology of the liver tissues in rats were observed after HE staining,and the protein expression of TLR-4,NF-κB,tumor necrosis factor- α(TNF- α) and interleukin- 2(IL- 2) in their liver was detected by Western blot method. Results: When compared with that in the control group, in the model group, there was disorderly arrangement of the liver cells, inflammatory cell infiltration and lipid droplets of different sizes;the protein expression of TLR-4,NF-κB,TNF-α and IL-2 was increased(P<0.05). When compared with those in the model group, in the groups of high- dose and low- dose, pathological morphology of the liver approached that in the control group; the arrangement of the liver cells was more orderly; inflammatory cell infiltration was significantly relieved; the number of small lipid droplets in the liver cells was decreased;cell vacuolization was significantly improved;the protein expression of TLR-4,NF-κB,TNF-α and IL-2 was decreased(P<0.05);the effect in the linderae aggregate group of high-dose was more significant. Conclusion:The extract of linderae aggregate leaf has significant improving effect on changes in pathology of the liver tissues in rat models with hyperlipidemia. Its mechanism is probably realized by blocking TLR- 4/NF- κB signaling pathway, and reducing protein expression of TNF-α and IL-2.
