    目的:探究应用HPLC-UV-DPPH 在线分析法评价炮制前、后地黄药材抗氧化活性成分的变化。方法:以毛蕊花糖苷为对照,选取4 批湖北、2 批安徽共6 批地黄作为样品,并以S01~06 编号, 选用Aglient Extend C18 色谱柱(250 mm×4.6 nm,5 μm),以乙腈-0.1%醋酸水溶液为流动相,进样20 μL,温度25 ℃、HPLC 检测波长334 nm、DPPH 检测波长517 nm,快速分析抗氧化活性效价;并采用HPLC-FT-MS 法(毛细管温度320 ℃,电压2.5 kV,电流100 μA) 进行成分分析。结果:以毛蕊花糖苷倒峰面积为对照,应用Y=0.0006X4-0.0241X3+0.4041X2+1.299X+1.652方程计算抗氧化性活性效价,以S02批次炮制后活性大幅度减少,S03、S04、S05 3 个批次活性成分大幅度增加,余批次差异不大。抗氧化成分比较,炮制后减少了1 种活性物质,但新增5种活性物质。结论:HPLC-UV-DPPH在线法分析地黄炮制前、后抗氧化活性,2种药材比较,既有新的抗氧化活性物质的生成,又有原抗氧化活性物质的消失。

    Abstract:

    Abstract: Objective: To investigate the changes of antioxidant active components of Rehmannia Glutinosa before and after processing by online HPLC- UV- DPPH method. Methods: Taking the mullein glycosides as control, four batches of Rehmannia Glutinosa from Hubei Province and two batches from Anhui Province were selected as samples with serial numbers of S01 to S06. By taking a mobile phase of acetonitrile- 0.1% acetic acid solution,sample introduction of 20 μL at 25℃ , and the wavelength detected by HPLC being 334 nm as well as the wavelength by DPPH being 517nm, the antioxidant activity was analyzed. The HPLC-FT-MS method(capillary temperature of 320 ℃,2.5 kV,100 μA) was used for component analysis. Results: The antioxidation activity titer was calculated by using the area of mullein antidrome as the control through the equation Y=0.0006X4-0.0241X3+0.4041X2+1.299X+1.652. After processing,the activity of Batch S02 was significantly decreased, while the active components of Batch S03, S04 and S05 were significantly increased, with little difference between the remaining batches. After processing,one active substance was reduced,but five active substances were added. Conclusion: Through an analysis of antioxidant activity of Radix Rehmanniae before and after processing by online HPLC-UV-DPPH method,there are both the formation of new antioxidant active substances and the disappearance of the original antioxidant active substances.
