    目的:观察针刺预处理对缺氧缺血性脑病 (HIE) 模型乳鼠血脑屏障 (BBB) 的保护作用,探讨针刺治疗 HIE 的作用机制。方法:将 70 只 SPF 级乳鼠随机分为假手术组、模型组、针刺预处理组。除假手术组外,采用结扎左侧颈总动脉然后进行 3.5 h 缺氧以建立 HIE 模型。采用悬吊实验评估实验动物的运动能力,苏木素-伊红(HE)染色观察神经细胞缺氧缺血后的病理学变化,透射电镜观察 BBB 亚显微结构,脑干湿重法测量含水量,蛋白免疫印迹法检测脑缺氧缺血后闭锁蛋白 (Occludin)、紧密连接蛋白-1 (ZO-1) 表达水平。结果:与假手术组比较,模型组乳鼠悬吊实验维持时间减少 (P<0.05),缺氧缺血侧脑组织含水量增加 (P<0.05),皮层组织 Occludin、ZO-1 蛋白表达下降 (P<0.05)。与模型组比较,针刺预处理组乳鼠悬吊实验维持时间增加 (P<0.05),缺氧缺血侧脑组织含水量减少 (P<0.05),皮层组织 Occludin、ZO-1 蛋白表达升高 (P<0.05)。结论:针刺可能通过上调 Occludin、ZO-1 蛋白表达维持 BBB 的完整性,从而改善缺氧缺血性脑损伤。

    Abstract:

    Abstract: Objective: To observe the protective effect of acupuncture preconditioning on blood- brain barrier(BBB) of neonatal rats with hypoxic ischemic encephalopathy(HIE),and to explore the mechanism of acupuncture for HIE. Methods:A total of 70 SPF neonatal rats were randomly divided into the sham operation group,the model group and the acupuncture preconditioning group. Except for the sham operation group,HIE models were established by ligating the left common carotid artery and keeping hypoxia for 3.5 hours. Sports ability of experimental animals was evaluated by hanging test;pathological changes in nerve cells after hypoxia- ischemia were observed by hematoxylin- eosin(HE) staining;BBB ultrastructure was observed by transmission electron microscope;water contents were measured by dry-wet method;protein expressions of Occludin and tight junction protein zona occludens-1(ZO-1) after cerebral hypoxia ischemia were detected by Western blot. Results:Compared with those in the sham operation group,the duration of hanging test in the model group was shortened (P<0.05);water content in hypoxic ischemic brain tissue was increased(P<0.05);the protein expressions of Occludin and ZO-1 in cortex tissue were decreased(P<0.05). Compared with those in the model group,the duration of hanging test in the acupuncture preconditioning group was increased(P<0.05);water content in hypoxic ischemic brain tissue was decreased (P<0.05);the protein expressions of Occludin and ZO-1 in cortex tissue were increased(P<0.05). Conclusion:Acupuncture can maintain BBB integrity by up- regulating protein expressions of Occludin and ZO-1,so as to improve hypoxic ischemic brain injury.
