Abstract: Objective: To identify the main natural saccharides in Huangqi Shengmai yin, and to determinate their contents. Methods: The main natural saccharides in Huangqi Shengmai yin were identified by comparing the peak time on high-performance liquid chromatography-evaporative light scattering detector(HPLC-ELSD) with that of standard reference substance. The content determination of monosaccharide was performed by COSMOSIL 5NH2-MS amino column combined with HPLC-ELSD;the mobile phase was acetonitrile-water(80:20);the flow rate was 1 mL/min;the column temperature was 35 ℃. Results:The main natural saccharide in Huangqi Shengmai yin was fructose. The HPLC-ELSD fructose detection method based on amino column showed good linear relationship in the concentration range of 0.2 to 16 mg/mL, with an average recovery rate of 98.63% and RSD of 1.76% . Conclusion: Huangqi Shengmai yin has relatively high content of fructose which can be detected via HPLC-ELSD method.
