Hewei Fan liukang Has Effect on Expression of IKKβin Gastric Mucosa of Rats with Bile Reflux Gastritis

目的:探究和胃反流康对胆汁反流性胃炎大鼠胃黏膜的作用机制。方法:通过灌胃反流液建立胆汁反流性胃炎大鼠模型,将40只大鼠随机平均分为空白组、模型组、磷酸铝凝胶组、和胃反流康组。通过Western blot法测定胃黏膜上皮IκB激酶复合体β(IKKβ)蛋白表达,通过RT-PCR法检测胃黏膜IKKβ表达含量。结果:与空白组比较,模型组大鼠胃黏膜组织IKKβ蛋白及IKKβmRNA表达水平显著升高( P <0.05)。与模型组比较,和胃反流康组、磷酸铝凝胶组大鼠胃黏膜组织IKKβ蛋白及IKKβmRNA表达水平显著降低( P <0.05, P <0.01)。与磷酸铝凝胶组比较,和胃反流康组无明显差异。结论:和胃反流康可能通过下调IKKβ基因的表达来有效地改善胃黏膜炎症。
Abstract:
Objective: To explore the mechanismof Hewei Fanliukang on the gastric mucosa of rats with bile reflux gastritis.Methods:Established rat models of bile reflux gastritis by gavage of reflux fluid.Forty rats were randomly divided into the blank group,the model group,the alumino phosphate gel group and the Hewei Fanliukang group.Detected the protein expressions of Iκ B kinaseβ (IKKβ)in gastric mucosalepithelium by Western blot,and IKKβ expression contents in gastric mucosa by RT-PCR.Results:Compared with the blank group,the expression levels of IKKβ protein and IKKβ m RNA in the gastric mucosa in the modelgroup were significantly increased( P <0.05).Compared with the modelgroup,the expression levels of IKKβ protein and IKKβ m RNA in the gastric mucosa of rats in the Hewei Fanliukang group and the alumino phosphate gel group were significantly decreased( P <0.05, P <0.01).Compared with the aluminumphosphate gel group,there was no significant difference being found in the Hewei Fanliukang group.Conclusion:Hewei Fanliukang may effectively improve gastric mucosal inflamm ation by reducing gene expression of IKKβ.
