    目的:探讨何首乌提取物对系膜增生性肾小球肾炎(MsPGN) 大鼠的治疗作用及对肾组织中核转录因子κB(NF-κB)表达的影响。方法:将40只Wistar大鼠按体质量随机分为对照组、模型组、何首乌提取组、洛汀新组4组,每组10只。除对照组外,其余各组进行单侧肾脏切除,连续多次注射OX7抗体诱导MsPGN大鼠模型,何首乌提取物组与洛丁新组于注射抗体4周后分别灌胃何首乌提取物和洛丁新治疗,对照组和模型组给予同等体积的生理盐水灌胃,治疗6 周后处死大鼠。测定模型制备前、注射抗体4 周后、治疗6 周后大鼠的24 h 尿蛋白定量水平;收集大鼠血清,测定血清中肌酐(SCr) 和尿素氮(BUN) 含量;肾脏切片后HE染色观察组织病理学改变;逆转录聚合酶链式反应(RT-PCR)检测NF-κB p65和单核细胞趋化蛋白-1(MCP-1)mRNA表达水平。结果:治疗6周后,与对照组比较,模型组大鼠24 h尿蛋白定量水平及血清中SCr、BUN含量显著升高,肾组织可见明显的系膜基质增生和系膜细胞增殖,NF-κB p65和MCP-1 mRNA表达水平显著升高,差异均有统计学意义(P<0.05)。与模型组比较,洛汀新组和何首乌提取物组大鼠24 h尿蛋白定量水平及血清中SCr、BUN含量显著降低,肾脏组织病理病变程度减轻,NF-κB p65和MCP-1 mRNA表达水平显著降低,差异均有统计学意义(P<0.05)。结论:何首乌提取物对MsPGN大鼠有一定的治疗作用,阻断NF-κB信号活化可能是其机制之一。

    Abstract:

    Abstract: Objective: To discuss the curative effect of extract of Radix Polygoni Multiflori on rats with mesangial proliferative glomerulonephritis(MsPGN) and its effect on the expression of nuclear factor kappa- B(NF- κB) in the kidney tissue. Methods: A total of 40 Wistar rats were randomly divided into the control group, the model group, the extract of Radix Polygoni Multiflori group and the Lotensin group according to their body mass,10 rats in each group. Except those in the control group,rats in the other groups were given renal resection on one side and continually injected with OX7 antibody to induce MsPGN rat models. After four-week injection,the extract of Radix Polygoni Multiflori group and the Lotensin group were respectively given extract of Radix Polygoni Multiflori and Lotensin by gavage, and the control group and the model group were given saline of the same volume by gavage. After six-week treatment,the rats were sacrificed. The levels of 24-hour urine protein were measured before establishment,after four-week injection and after six-week treatment. The ratserum was collected,and the contents of serum creatinine(SCr) and blood urea nitrogen(BUN) were measured. After renal section and HE staining,the pathological changes of the kidney were observed. The expression levels of mRNA of NF- κB p65 and monocyte chemoattractant protein- 1(MCP- 1) were detected by reverse transcriptase polymerase chain reaction (RT-PCR). Results:After six-week treatment,compared with those in the control group,in the model group,the level of 24- hour urine protein and the contents of SCr and BUN in serum were significantly increased; there were significant proliferation of mesangial matrix and cells in the kidney tissue;the expression levels of mRNA of NF- κB p65 and MCP- 1 were significantly increased;differences being significant(P<0.05). Compared with those in the model group,in the extract of Radix Polygoni Multiflori group and the Lotensin group,the levels of 24- hour urine protein and the contents of SCr and BUN in serum were significantly decreased; the degree of pathological changes of the kidney tissue was decreased; the expression levels of mRNA of NF- κB p65 and MCP- 1 were significantly decreased;differences being significant(P<0.05).Conclusion: Extract of Radix Polygoni Multiflori has certain curative effect on MsPGN rats, and its interruption on the activation of NF-κB may be one of its mechanisms.
