    目的:通过网络药理学方法研究黄芩治疗艾滋病的作用机制。方法:在线检索黄芩的化学成分、作用靶点及艾滋病的疾病基因,建立蛋白互作网络。通过在线DAVID 进行GO 功能富集分析和KEGG 通路富集分析,明确黄芩治疗艾滋病的生物学过程及信号通路。结果:收集到黄芩36个高效化合物、110靶点,与艾滋病的1781个疾病靶点进行基因映射,得到48个映射基因;构建蛋白互作网络,得到4个关键的化合物:金合欢素、汉黄芩素、三羟黄酮、β-谷甾醇,3个关键靶点:Bax、Bcl2、CASP8。GO 功能分析显示,艾滋病的基因功能主要是炎症应答、细胞质、染色质结合等方面。KEGG 信号通路富集分析得到肿瘤信号通路、p53 信号通路、PI3K-AKT 信号通路等条目。结论:通过网络药理学分析,中药黄芩可能通过其所含金合欢素、汉黄芩素、三羟黄酮、β-谷甾醇,作用于肿瘤信号通路、p53 信号通路、PI3K-AKT 信号通路的Bax、Bcl2、CASP8 靶点,调控炎症应答、细胞质、染色质结合等达到治疗艾滋病的作用。

    Abstract:

    Abstract: Objective: To study the mechanism of Radix Scutellariae in treating AIDS through network pharmacology. Methods:The chemical constituents and targets of Radix Scutellariae as well as the genes of AIDS were searched online to establish a protein interaction network. The biological process and signal pathway of Radix Scutellariae in treating AIDS were identified by GO function enrichment analysis and KEGG pathway enrichment analysis through online DAVID. Results:A total of 36 effective constituents and 110 targets of Radix Scutellariae were obtained,and they were mapped to 1,781 disease targets of AIDS,and then 48 mapped genes were obtained. Four key chemical constituents were obtained by constructing the protein interaction network, namely, acacia, baicalin, trihydroxyflavone and β- sitosterol, and three key targets, namely, Bax, Bcl2 and CASP8. GO function analysis showed that the gene functions of AIDS were mainly inflammatory response, cytoplasm and chromatin binding. By KEGG pathway enrichment analysis, tumor signaling pathway, p53 signaling pathway and PI3K-AKT signaling pathway were obtained. Conclusion:Through the analysis of network pharmacology, Radix Scutellariae may affect Bax,Bcl2,CASP8 targets of tumor signaling pathway,p53 signaling pathway and PI3K AKT signaling pathways through acacia , baicalin , trihydroxyflavone and β- sitosterol , to regulate inflammatory response , cytoplasm,and chromatin binding to treat AIDS.
