    摘要:

    目的:研究珍斛颗粒对异丙肾上腺素(Isoproterenol,ISO)诱导大鼠心肌缺血损伤的保护作用。方法:32只清洁级健康SD大鼠随机分为空白组、模型组、低剂量组、高剂量组,每组8只,先适应性喂养7天。低、高剂量组予珍斛颗粒按大鼠体表面积换算量药液灌胃,空白组和模型组给予同容积生理盐水,每天1次,连续14天。末次灌胃30 min后,麻醉各组大鼠记录Ⅱ导联心电图,模型组,低、高剂量组皮下注射ISO复制心肌缺血模型,空白组给予同容积生理盐水,每天1次,连续2天,分别记录30 min和末次皮下注射ISO后12 h的心电图,观察心电图J点变化。描记心电图后,将各组大鼠从腹主动脉取血,离心后检测血清中血清肌酸激酶(Creatine kinase,CK)、肌酸激酶同工酶(Creatine kinase isoenzyme,CK-MB)、乳酸脱氢酶(Lactate dehydrogenase,LDH)水平,并取各组大鼠心尖部心肌组织,进行HE染色,观察心肌缺血损伤程度。结果:与空白组比较,模型组注射ISO 30 min后及末次皮下注射ISO 12 h后,心电图J点位移程度显著升高(P<0.05);血清CK、CK-MB、LDH均显著增加(P<0.05)。与模型组比较,珍斛颗粒高、低剂量组注射ISO30 min后及末次皮下注射ISO12 h后心电图J点位移程度显著降低(P<0.05);血清CK、CK-MB、LDH均明显降低(P<0.05);低、高剂量组间无显著性差异。HE染色显示模型组心肌细胞明显损伤,低、高剂量组心肌细胞损伤较模型组减轻。结论:ISO可引起大鼠心电图出现缺血改变,心肌酶谱升高,导致心肌缺血损伤,珍斛颗粒对缺血心肌有明显保护作用。

    Abstract:

    To study the protective effect of Zhenhu granules for myocardial ischemia injury induced by isoproterenol(ISO)in rats.Methods:Divided 32 healthy SD rats into the blank group,the model group,the low dose group and the high dose group randomly,with 8 cases in each group,and adaptively fed them for 7 days.The low and high dose groups were given Zhenhu granules by gavage according to the conversion amount of the body surface area of rats,and the blank group and model group were given normal saline of the same volume once a day for 14 consecutive days.30 min after the last gastric perfusion,recorded II lead electrocardiogram of anesthetized rats in each group;the model group,the low and high dose groups were given subcutaneous injection of ISO and established the myocardial ischemia model.The blank group was given normal saline of the same volume once a day for 2 consecutive days.Respectively recorded the electrocardiogram 30 min later and 12 h after the last subcutaneous injection of ISO,observed the changes of J point of electrocardiogram.After recording the electrocardiogram collected the blood from the abdominal aorta of rats in each group,detected the levels of serum creatine kinase(CK)creatine kinase isoenzyme(CK-MB)and lactate dehydrogenas(LDH)after centrifugation and took myocardium of heart apex of rats in each group for HE staining and observed the degree of myocardial ischemia damage.Results:Compared with that in the blank group,the displacement of J point in the model group significantly was increased in 30 min after ISO injection and in 12 h after the last subcutaneous injection of ISO(P < 0.05);serum CK,CK-MB and LDH were all significantly increased(P<0.05).Compared with that in the model group,the displacement of J point of the high and low dose groups of Zhenhu granules was significantly reduced in 30 min after ISO injection and in 12 h after the last subcutaneous injection of ISO(P < 0.05);serum CK,CK-MB and LDH were all significantly reduced(P < 0.05);there was no significant difference being found in the comparison between the low and high dose groups.HE staining indicated that there was apparent damage on myocardial cells in the model group,while the damage of myocardial cells in the low and high dose groups was reduced when compared with that in the model group.Conclusion:ISO can induce ischemic changes in the electrocardiogram of rats,increase the myocardial enzymes and lead to myocardial ischemia injury.The application of Zhenhu granules has obvious protective effect on ischemic myocardium.
