    目的:探讨电针天枢穴对溃疡性结肠炎(Ulcertive colitis,UC)大鼠的疗效及其对TLR4/NF-κB信号通路的影响。方法:75只Wistar大鼠随机分为空白组、模型组、电针组、假电针组和柳氮磺胺吡啶组(SASP组),除空白组外均采用2,4,6三硝基苯磺酸/乙醇溶液灌肠复制UC大鼠模型。电针组给予电针双侧天枢穴,假电针组给予电针假天枢穴(天枢穴旁开1寸),SASP组给予10 mL/kg柳氮磺胺吡啶悬浊液灌胃治疗。连续治疗14天后观察各组大鼠DAI评分,光镜下观察结肠组织病理学,ELISA法检测血清白细胞介素-1β(Interleuki-1,IL-1β)、肿瘤坏死因子-α(Tumor Necrosis Factor,TNF-α) 及白细胞介素-10(Interleuki-10,IL-10) 的含量,Western-blot及RT-PCR检测结肠组织中Toll样受体4(Toll-like receptors,TLR4)、髓样分化初级反应 88(Myeloid differentiation primary response 88,MyD88)、核因子 κB p65(Nuclear factor kappa-light-chain-enhancer of activated B cells p65,NF-κB p65)蛋白和mRNA的表达。结果:与空白组比较,模型组大鼠DAI评分及血清IL-1β、TNF-α含量显著增高,IL-10含量显著降低,结肠组织中TLR4、MyD88、NF-κB p65的表达均显著增高(P<0.05);与模型组比较,电针组大鼠DAI评分显著降低,血清IL-1β、TNF-α含量显著降低,IL-10含量显著增高,结肠组织中TLR4、MyD88、NF-κB p65的表达均显著降低(P<0.05);与SASP组比较,电针组DAI评分及血清IL-1β含量较低、IL-10含量较高,结肠组织中TLR4、MyD88、NF-κB p65蛋白均较低(P<0.05)。结论:电针天枢穴能够明显改善UC大鼠的结肠组织病理学形态,降低DAI评分,治疗UC,其作用机制是通过抑制TLR4/NF-κB信号通路的活化实现的。

    Abstract:

    Objective:To observe the therapeutic effect of the therapy of electroacupuncture on Tianshu(ST25)for rats with ulcerative colitison(UC)and its effect on the TLR4/NF-κB signaling pathway.Methods:Randomly divided 75 Wistar rats into the blank group, the model group, the electroacupuncture group, the sham electroacupuncture group and salazinsulfonapyridine(SASP)group.In addition to the blank group,the UC rat models in other groups were replicated by 2,4,6 trinitrobenzene sulfonic acid/ethanol solution by enema.The electroacupuncture group received the electroacupuncture on Tianshu(ST25)on both sides.The sham electroacupuncture group received the electroacupuncture on the shame Tianshu(ST25)where located on one inch beside it.The SASP group was treated with 10 mL/kg SASP suspension by gavage.After 14-day treatment,evaluated DAI scores,observed the pathology of colon tissues under the light microscope,and detected the contents of interleukin-1β(IL-1β)in serum,tumor necrosis factor-α(TNF-α)and interleukin-10(IL-10)by ELISA method,as well as observed the expression of protein and mRNA in toll-like receptors4(TLR4),myeloid differentiation primary response 88(MyD88)and nuclear factor kappa-light-chain-enhancer of activated B cells p65(NF-κB p65)in colon tissues in all groups by Western-blot and RT-PCR.Results:Compared with the blank group,the DAI score and contents of IL-1β and TNF-α in serum in the model group were significantly increased, the content of IL-10 was significantly decreased,and the expression of TLR4,MyD88,and NF-κB p65 in colon tissues was significantly increased(P<0.05).Compared with the model group,the DAI score in the electroacupuncture group and contents of IL-1β and TNF-α in serum were significantly decreased,the content of IL-10 was significantly increased,and the expression of TLR4,MyD88,and NF-κB p65 in colon tissues was significantly decreased(P<0.05).Compared with the SASP group,the DAI score in the electroacupuncture group was and content of IL-1β was lower and IL-10 was higher;protein expression of TLR4,MyD88,and NF-κB p65 in the colon tissues was lower(P<0.05).Conclusion:The therapy of electroacupuncture on Tianshu(ST25)can significantly improve the pathological morphology of colon tissues in rats with UC,reduce DAI score and treat UC.Its mechanism is to inhibit the activation of TLR4/NF-κB signaling pathway.
