    摘要:

    目的:观察盆舒颗粒对宫腔黏连模型大鼠子宫内膜转化生长因子-β1(TGF-β1)、信号传导蛋白(Smad3) 的影响。方法:将50只成年雌性SD大鼠随机分为正常组、模型组、中药治疗组、TGF-β1抗体干预组、TGF-β1抗体干预+中药治疗组,每组10只,采用自制刮宫勺刮伤大鼠子宫内膜制作宫腔黏连模型,各组分别给予相应的干预措施。在干预第15天摘取双侧子宫,观察各组大鼠宫腔组织损伤病变情况,并采用免疫组化法检测各组大鼠子宫组织中TGF-β1,Smad3含量。结果:模型组与正常组95%可信区间无重叠,说明有显著性差异;模型组与中药治疗组、TGF-β1抗体干预组、TGF-β1抗体干预+中药治疗组95%可信区间均无重叠,说明有显著性差异;中药治疗组与TGF-β1抗体干预组、TGF-β1抗体干预+中药治疗组95%可信区间重叠,说明无显著性差异。TGF-β1抗体干预+中药治疗组疗效最好。与正常组比较,模型组大鼠子宫内膜上皮细胞TGF-β1、Smad3表达显著升高,差异均有统计学意义( P <0.05)。与模型组比较,中药治疗组、TGF-β1抗体干预组、TGF-β1抗体干预+中药治疗组大鼠子宫内膜上皮细胞TGF-β1、Smad3表达显著降低,差异均有统计学意义( P <0.05)。中药治疗组、TGF-β1抗体干预组、TGF-β1抗体干预+中药治疗组3组大鼠子宫内膜上皮细胞TGF-β1、Smad3表达之间比较,差异无统计学意义(P>0.05)。结论:盆舒颗粒、TGF-β1抗体能改善宫腔黏连模型大鼠子宫病理变化,降低TGF-β1、Smad3表达,具有潜在防治宫腔黏连的作用,二者联合使用作用效果更为显著。

    Abstract:

    Objective:To observe the effect of Penshu granules on transforming growth factor-β1(TGF-β1)and signal transduction proteins(Smad3)of endo metriumin model rats with intrauterine adhesion.Methods:Divided 50 cases of adult female SD rats into the normal group,the model group,the Chinese medicine treatment group,the TGF-β1 antibody intervention group and the TGF-β1 antibody intervention plus Chinese medicine treatment group randomly,10 cases in each group.After establishing model of intrauterine adhesion by using self-made curettage spoon to scratch the endo metriumin model rats.Each group was given corresponding intervention,the bilateral uterus was taken to observe the lesion of uterine tissues of rats in each group on the 15 th day of treatment,and the contents of TGF-β1 and Smad3 of rats in each group were detected by immunohis to chemical method.Results:The 95%confidence interval between the model group and the normal group was not overlapped,indicating significant difference.The 95%confidence interval between the model group and the Chinese medicine treatment group,the TGF-β1 antibody intervention group or the TGF-β1 antibody intervention plus Chinese medicine treatment group was not overlapped,indicating significant difference.The 95%confidence interval between the Chinese medicine treatment group and the TGF-β1 antibody intervention group or the TGF-β1 antibody intervention plus Chinese medicine treatment group was overlapped,indicating no significant difference.The curative effect in the TGF-β1 antibody intervention plus Chinese medicine treatment group was the best.Compared with the normalgroup,the expression of TGF-β1 and Smad3 of epithelial cells in endo metriumwas significantly increased in the model group,differences being significant(P < 0.05).Compared with the model group, the expression of TGF-β1 and Smad3 of epithelial cells in end ometrium was significantly decreased in the Chinese medicine treatment group,the TGF-β1 antibody intervention group and the TGF-β1 antibody intervention plus Chinese medicine treatment group,differences being significant(P < 0.05).No significant difference was found in the comparisons of expression of TGF-β1 and Smad3 between the the Chinese medicine treatment group,the TGF-β1 antibody intervention group and the TGF-β1 antibody intervention plus Chinese medicine treatment group(P > 0.05).Conclusion:Penshu granules and TGF-β1 antibody can improve the pathological changes ofmodel rats with intrauterine adhesion, reduce the expression of TGF-β1 and Smad3, and have the potential to control the intrauterine adhesion.The effect of the combination of the two is the more significant.
