目的:研究徐氏化纤汤对四氯化碳(CCl4)诱导的肝纤维化大鼠的作用及其可能的作用机制。方法:将72只SD大鼠随机分为空白对照组、模型组、化纤汤高剂量组、化纤汤中剂量组、化纤汤低剂量组、秋水仙碱组,每组12只。除空白对照组外,其他各组均采用40% CCl4+花生油腹腔注射制作大鼠肝纤维化模型。空白对照组和模型组每天灌服生理盐水;化纤汤高、中、低剂量组每天分别灌服21 g/(kg·d)、10.5 g/(kg·d)、5.25 g/(kg·d)化纤汤药液;秋水仙碱组按0.35 mg/(kg·d)灌服秋水仙碱药液。检测各组大鼠肝功能、肝纤四项,并作肝组织病理切片分析反映肝脏的损伤程度。结果:与空白对照组比较,模型组大鼠肝重、肝脏指数明显升高,大鼠体质量明显减轻,血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TBil)、透明质酸(HA)、Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(CⅣ)显著升高,血清白蛋白(Alb)显著降低,差异均有统计学意义(P<0.05)。与模型组比较,秋水仙碱组、化纤汤各剂量组大鼠体质量明显升高,大鼠肝重、肝脏指数明显降低,大鼠血清ALT、AST、TBil、HA、CⅣ、PCⅢ显著降低,Alb显著升高,差异均有统计学意义(P<0.05)。结论:徐氏化纤汤有一定的保护肝功能及抗肝纤维化的作用。 |

Abstract:Objective:To study the effect of Xu's Huaxian tang on rats with hepatic fibrosis induced by carbon tetrachloride(CCl4)and its possible mechanism.Methods:Divided 72 SD rats into blank control group,model group,Huaxian tang high dosage group,Huaxian tang middle dosage group,Huaxian tang low dosage group,colchicine group randomly,12 rats in each group.Except blank control group,all the other groups received intraperitoneal injection of 40%CCl4with peanut oil to establish the hepatic fibrosis rat model.Blank control group and model group were gavaged with normal saline every day;Huaxian tang high,middle and low dosage groups were respectively gavaged with 21 g/(kg·d),10.59 g/(kg·d)and 5.25 g/(kg·d)Huaxian tang every day;colchicine group was gavaged with 0.35 mg/(kg·d)colchicine.Detected the liver function and four indicators of hepatic fibrosis in each group,and analyzed the damage degree of liver by making pathological section of liver tissue.Results:Comparing with those in blank control group,in the model group,the liver weight and liver indexes were obviously increased,while the body mass was significantly reduced;serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBil),hyaluronic acid(HA),procollagen type Ⅲ(PCⅢ),and collagen Type Ⅳ(CIV)were all markedly risen,while serum levels of albumin(Alb)were evidently decreased,differences all being significant(P<0.05).Comparing with those in model group,in colchicine group and Huaxian tang high,middle and low dosage groups,body mass was increased obviously,the liver weight and liver indexes were obviously reduced,levels of ALT,AST,TBil,HA,CIV,and PCⅢ were all evidently decrease,and levels of Alb were significantly increased,differences all being significant(P<0.05).Conclusion:Xu's Huaxian tang has certain effect on protecting liver function and treating hepatic fibrosis. |
